Evaluating transformation

Colonies were picked for all the antisense RNA constructs and were evaluated on a gel. with the result that all constructs were present in some colonies carrying the correct construct except E1B3.


Triparental mating

The conjugation plasmi carrying strain was inoculated in preparation for triparental mating.


Transformation

Yesterdays digestions of RPRAT, RPRYT, PRKRYT were transformed. pPetE + RPRAT, pPetE + RPRYT and PRKRYT were moved to AC in preparation of triparental mating, since the carrier plasmid in only available with kanamycin resistance.
