Colony PCR of the most recent transformations

A colony PCR was performed with Phusion for the constructs: RBS-PirA-RBS-PirB-T, RBS-pdc-RBS-ADHZ-T. Only one of these the (BS-pdc-RBS-ADHZ-T (4)) actually showed the correct length.

This was inoculated at 16:20.

Transformations

Transformations performed: kivd + RBS-YADH2-T to AK3, pPetE-RBS to AK3, RBS + kivd to AC3, RBS-PirA + Rbs-PirB-T to AK3, RBS-pdc + RBS-ADH2Z to AC3 and kivd in the puc19 plasmid it was delivered in.

The ligations were plated at 19:30

Colony PCR of antisense RNA

To better evaluate the results from the antisense RNA constructs a new PCRwas performed. The rsult showed that pPetE-E1A2:1-T, pPetE-E1A3:2-T, pPetE-E1A4:1-T, pPetE-E1B2:2-T, pPetE-E1B4:1-T, pPetE-E1B4:2-T, pPetE-E22:1-T, pPetE-E22:2-T, pPetE-E23:1-T, pPetE-E23:2-T and pPetE-E24:1-T seemed to have worked and these were inoculated overnight.
