Preparing competent cells
Continued since yesterday
The seed stock that was inoculated in 300 ml SOB yesterday was far to dense when the OD600 was measured 15h after inoculation. Because of this we prepared a new flask and do the inoculation in the morning instead, to be able to monitor the growth.
Tansforming ADH2 Z.mobilis
Continued since yesterday
A plasmid prep was performed with quite low concentrations of DNA so this will be redone tomorrow as well.
Glycerol stock of ADH2 z
Glycerol stocks (15%) of ADH2 from Z. mobilis was prepared an put in the freezer. --Flormane 14:29, 20 August 2009 (UTC)
ADH2 Z.mobilis Inoculation
I inoculated 7.5ml of LB with the glycerol stocks of ADH2 (Z) on pSB1A3 in E. coli tonight for the upcoming MiniPrep tomorrow.
