The pPetE PCR product were digested with EcoRI + PstI and ligated into psb1A3. After that the ligated plasmid were transformed into Top10 competent cells through heat-shock for 55 sec in 42 deg C and incubated over night.

Picking colonies

Colonies were picked from the transformations made yesterday.
