Colony PCR and Gel evalutation
Gel of the PCR from yesterday evening was performed. Actually we found out that the new thiozole orange buffer for the gel was faulty and caused the ugly bands. New TO Buffer was preparred accroing protocol.
Making competent cells
continued since many days :)
Today we harvested the culture from yesterday, we transfered the cells to 2 ml tubes with 100 µl of cells in each. The cells will be transformed today and evaluated tomorrow.
Mini prep
Plasmid preps were performed on the cultures carrying the PBS+PirB and PBS+ADH2. (Mini Prep Kit by Sigma)
