Competent cell preperation

We made CCMB80 buffer and inoculated four 2 ml cultures of competent E.coli as given by: [1]. Depatures from protocol in this step was the use of fake 'SOB' media, we used crancked up LB, and also the autoclavation of the CCMB80 buffer as opposed to the recommended sterile filtration. this depature from the protocol left os with a brownish percipitation in the buffer, we hope this won't affect the buffer to much.
