Making LB (Broth)
1. Add 20g LB into 2. 1 L ddH2O in 2L of Flask ➢ Swirl the flask until all the LB is dissolved. ➢ Aliquot them into smaller volume (usually into 100ml bottles). ➢ Autoclave (at 20min at 15psi and 121 oC) them with cap loose but covered with aluminum foil. • Tighten the caps and leave the medium bottle without any antibiotics in it. • When it is about to be used, the person can add the selected antibiotic (the right concentration and the right volume): o Ampicillin (Amp) = 1ul of [50ug/ml] for every 1ml of medium. o Kanamycin (Kan) = 1 ml of [50mg/ml] for every 1ml of medium. o Tetracycline (Tet) = 1 ml of [12.50mg/ml] for every 1ml of medium. o Chloramphenicol (Ch) = 1ml of [10mg/ml] for every 1ml of medium.
