Wet Lab
- Results of the RO1:
There is clones on every plate!
- → colony PCR on all clones with the Taq Platinium protocol
- Results from the media try:
All media have worked! (except for the SC) They have different colors depending on media and clone after an overnight culture.

We tried the following media, with and without Trp :
- LB
- M9/min + thiamine
- M9/min + AA + glycerol
- M9/min + thiamine
The gel showed :
- RO1: it works fine, all our constructs have the TrpO
- RO2: #6 and #7 do not have the construct (we took the wrong clones), while #8 seems to have the construct!
