Making SOB Medium ( For growing competent E.Coli cells)
• Making 1L of SOB • Please look at Pg 1356 of the Lab Reference Book for details. 1. 20g Bacto tryptone 2. 5 g of Bacto yeast Extract 3. 0.584g NaCl 4. 0.186g KCl 5. 1L ddH2O 6. 2.4g MgSO4 (OR MgCl2 in 90ml ddH2O ) ➢ Swirl the flask until the ingredients are completely dissolved. ➢ Autoclave the medium.
