Wet Lab

Results of yesterday transformation

LR2 + GFP grew (few colonies), RO1 didn't grow and RO2 + BB grew (few colonies). They are all single plasmid !!

Colony PCR

We did a colony PCR of the following bacteria :

6 clones of RO2 + BB5.

6 clones of GFP + LR2.

1 of each type of double transformant colonies (second try given the migration result of yesterday) : RO2#8/BB5, RO2#10/BB6, RO2#4/BB1, RO2#5/BB5.

Gel

We did a migration of the products from yesterday Taq platinium PCR protocol (RFP, Klenow, LR2, BB5 etc). They were run on a gel again because the bands seemed strange. Finally, the results were good except for the Klenow so we will do another gel for Klenow.

Culture

3ml of overnight culture has been made for the following clones :

1x LR2 + GFP 1

1x LR2 + GFP 2

1x LR2 + GFP 4

1x RO2#4 (in LB and in M9)

1x RO2#5 (in LB and in M9)

1x RO2#10 (in LB and in M9)

All these cultures will be cultivated tomorrow morning in 25 ml erlenmeier flasks for testing in Sebastian's lab.
