Wet Lab
Miniprep of the yesterday's liquid cultures to isolate the plasmids.
We tested the ligation protocol again this time deciding to check every detail of the process (concentrations, controls etc). We left the ligations overnight at 4°C.
Prepared primer mixes for the new IDT primers (for the 1.5-step PCR) and ran the 1.5-step PCR.
Transformation of the ligation products of the previous day, bacteria left to grow overnight at 37°C.
