Wet Lab
Miniprep of the clones which contained our 2nd read-out system.
Synthesis of the Trp operon sequence with Klenow fragment extension, purification, digestion, purification again and ligation during 1 hour at RT. Then, transformation of DH5-alpha cells with the ligation products, plates incubated overnight at 37°C.
Ligation during 1 hour at RT with the backbones obtained by gel extraction yesterday evening.
Transformation of DH5-alpha cells with a whole bunch of ligation products: LovTap, LovTap-Term, LacI-RBS-LovTap-Term, LacLP-RBS-LovTap-Term, T7-RBS-LovTap-Term, read-out 2 (total of 28 plates!!!!).
Plates left to incubate overnight at 37°C.
