Wet Lab
Colonies grew for the ligation LOVTAP-term !!!!!!! But not for the LacI-RBS ligation.
So colony PCR have been done to check if the plasmid contain the correct part (LOVTAP-term)
Miniprep for RBS was done.
After the colony PCR, we checked the PCR products with an agarose gel. Result: for one of the clones there was a band of the desired length! The corresponding clone was put into liquid culture to grow overnight at 37°C.
We also grew more LacI and death cassette plasmids by taking cells from the glycerol stocks since we didn't have much left in plasmids.
