12th August:
1. Induction of fosmid has been done with L-arabinose to increase the copy number.
2. Induction of GB05MN cells with L-rhamnose has been done to increase the copy number.
13th August:
1. Glycerol stocks of GB05MN, EPI300, GB05(pccFos-MeCP2-Bsd), GB05MN(pccFos-MeCP2-Gent-rpsL) are made.
2. Clones of pccFos-MeCP2-Bsd and pccFos-MeCP2-Gent-rpsLare inoculated in LB-Cm10 with L-arabinose for induction overnight.
14th August:
1. DNA isolation of pccFosMeCP2-Bsd and pccFosMeCP2-Gent-rpsL have been done.
2. Sample DNA from both preparations are run on a gel for checking.
