Wet Lab
A new gel was made to assess if the double transformants really weren't (the last gel was a bit fuzzy). 1% 100ml (done yesterday, stayed in TBE buffer overnight), so 190 V for 30 min. The result isn't better than yesterday.
Put RO1.1 and RO2.4 transformants in DH5alpha, RO1.1 or RO2.4 with mutated BB ( ILE427PHE) double transformants in DH5alpha to grow in 5ml LB
