  • The terminator colonies didn’t grow in the liquid LB medium.
  • We performed miniprep of the RBS inocule (Protocol 2).

Lux S (BBa_K091109) enzymatic digestion

  • We tried one more time the enzymatic digestion with XbaI and PstI, but didn’t find the part again. We expected a 530 band size, but we just noticed a plasmid band. Something is wrong with this part.
