I) DISCUSSIONS WITH PRAVEEN SIR...

  • Ligation needs to be practised.
  • PCR-ligation-Transformation takes time to come. no colonies even after 16 hours.
  • Troubleshooting,
  • Kan too High? Try lowering KAn.
  • Ligation, try blunt end with the cohesive
  • Is PCR the right way to manna? Try transformation.
  • Proceed with the current thing, wait patiently for colonies to appear.

II)Plasmid isolation started from single colony from P4N.
