Finally standardized the PCR for a part of the competence gene regulon. Com EF from Acinetobacter baylyi was successfully amplified and the post PCR cleanup was done.
A 16 hr TA ligation reaction set up today to ligate the amplicon into standard pGEMT and pCR2.1 vectors.
