I) Got Transformants. Inoculated them in Broth. Isolation Failed. x-(

II) Took a-MF >put a non specific PCR with Vf2 and Vr2

>Gel

>Got Band around 1.3

>Partial amp occurs.

>proceed with P2f P2r

>Same size amplified.

Conclusion-

PCR does not work to amplify BioBricks.

We have to amplify using TSS Protocol
