Primers
We have eluted all the Yeastguard project primers.
PCR with DNA from Kluyveromices lactis
We did the PCR reactions to amplificate the K. lactis sequences. We made a 20µL standardt reaction, using 0,8µM of each primer. The annealing temperatures were: 59ºC for DLD promoter; 57ºC for JEN1 promoter; 53ºC for JEN1 gene (agarose gel above).
Recovering biobrick: terminator
We plated the terminator transformation in LB without antibiotic, because we could´t find cells when we plated on LB+Amp or LB+Kan.
