Genomic DNA Extraction for Hly Operon Manipulation
We performed the genomic DNA Extraction from four pathogenic E. coli hly+ strains. We used the Aubusel 1998 protocol with modifications, Protocol 1 (see protocols section).

5 plates from yesterday transformation had colonies. The other 2 transformed bacteria were re-plated in two diferents antibiotics and in higher cell concentration, in the morning. At night one of these plates showed colonies. Overnight liquid cultures were grown from the single colonies that were transformed yesterday, each one contaning a diferent biobrick.
