YeastGuard

With the product of the mini-prep played yesterday we performed a PCR to verify the correct ligation of the ……… part into the ……. plasmid. In this PCR we used primers that join in the plasmid (VFoward and VReverse) and a primer that join in the insert (…… reverse).

Cre-REcombinase - transformations results

  • Today we checked yesterday's plates and, unfortunately, all grown colonies showed a red coloration!
  • That means that the vector probably religated with it's own part (RFP device). This could be avoided by purification of the plasmid's digested band.
