We tested a ligation with LacI-RBS without Phosphatase and with different concentrations of vector and insert. We also tried ligating a digested GFP plasmid. The ligations were left overnight.
We also made liquid cultures of RBS, GFP, LacI, backbones 1 and 2 (resistances to kanamycin and chlorophenicol).
