Miniprep, digestion and ligation of RBS

  • Today we perfomed miniprep from the RBS transformed E. coli according to the Protocol 2. The fragment size was correct and we decided to digest it according to the Protocol 14 with the SpeI and PstI enzymes. It worked.
  • After the RBS plasmid digestion, we ligated it to CFP (the reporter gene) according to the Protocol 11.
