Wet Lab
LacI, RBS and the death cassette were digested with E/S, X/P and E/P respectively. (Incubated at 37°C for 1h30) The digestion products have been purified, and than ligated (incubation at RT for 3 hours).
The digestion products have been runned on a gel to see if the digestion worked, it worked for the death cassette but we are not able to see the LacI bands and the RBS bands.
LOVTAP-term ligation products and LacI-RBS ligation products have been transformed in DH5-alpha.
Finally RBS plasmid has been retransformed to have RBS part again.
