Wet Lab

Miniprep of RBS and our LacI-RBS

Colony PCR of different LovTap-Term and digestion assay by Pst1. Result after an agarose gel: All colonies seem to have the LovTap part in a plasmid (the LovTap has two P sites).

We re-did a 1.5 step PCR of the T7, LacLP and LacI biobricks and keep the products for the next day.

We did a PCR of T7-CBP-LovTap-Term, a digestion, a ligation and transformation into a E1010 backbone.
