I helped Daniel making some plates, as well as made some more LB medium.

Today I minipreped the Riboregulator Assemblies 5,6,7, 8 (Check 24th August Lab notebook) along with the BBa_S03335 in Chloramphenicol backbone. Then I restricted the samples and ran a gel. Since the staining needed overnight check the image in tomorrow's notebook.

8 colonies of the transformation !Z( 2 from each plate)were grown overnight for miniprep. The used colonies are named:

Restriction with E and S of PCR results for Locks and Keys. Besides restriction with the same enzymes of pSB1A3 backbone. Ligation of locks and keys in Amp backbones. Transformation and culture in plates and tubes.

Got sequencing results back. CE sequence confirmed and CB partially confirmed. Other reactions did not work.
