Brain work
We encountered difficulty to begin the experiments in the morning , because we still lack lab furniture.
Real work started in the afternoon, while two of us participated to the "festival des sciences" event.
Unluckily no one comes and we started the conference
just between the present scientists and us. The debate tackle a lot of ethic problem, like
what are the responsability of he scientists involved in a work that is an extension of previous one ?
Is it because we can that we should ?
Lab work
- Glycerol stock for S1 DH5alpha and S2 K12 MG 1655 delta recA::Kan
- 0,66ml of 60% glycerol into cryogenic tube
- 1,32ml of overnight bacterial culture
- Vortex gently
- Stock at -80°C
- Culture of S1 DH5alpha if 5ml of LB
- Genomic DNA Purification with Wizard® Genomc DNA purification Kit of Promega
- Step 2: 14000rpm 2min
- Step 7: Cooling 10min
- Step 9: 45min 37°C
- Step 12: 14000rpm 3min
- Step 19: 1h 65°C, gently tapping the tube all the 10min
