We found why the sequencing failed last time. First of all Tim gave wrong primers secondly my stupidity I added both primers in same sample misinterpreting what Tim meant for two separate same larger DNA samples. So today I sent samples again for sequencing. I got nice colonies with the lacI generator attached. None of the colonies are now glowing. Hence I think my assembly worked.
