Today I checked in gel (see below) that the small colonies of LacI generator and Normal RBS control has the plasmids of correct sizes. Hence with the correct LacI generator I ligated the control plasmids, Plasmid 1 of negative cascade and locks and keys. Then they were electroporated to cells.
Tim Weenink
Ligated the following constructs: !Y and !X and transformed the E. Coli K12 strain with them.
