Today I was able to do miniprep alone of the LacI generator and Normal RBS Control since I had meeting today.

None of yesterdays inoculations showed GFP expression. All aTc induced inoculations should have. So I did PCR reactions to check the lengths of the inserts.
These reactions were gelled:
After that I did replicaplating of all !Z plates on plates with Cam and Cam&aTc. This way I could screen for colonies that did express GFP when induced with aTc. But it later turned out that none of the colonies on the plate with aTc expressed GFP.
