Creation of the cro-box testing devices

• Colonies selection
• R0010 brick isolation

Methods:

• Appropriate colonies carrying a cro-box+reporter device were selected (5 for each variant) and transferred to a new plate. Liquid cultures were established along the way. (in LB supplemented with ampicillin).
• Appropriate colonies carrying the I14018 brick were selected and transferred to a new plate. Liquid cultures were established along the way.(in non-supplemented LB medium).
• The R0010 birck was isolated 4 times from 3μl of overnight culture using the Plasmid Mini kit (A&A Biotechnology).
• The transfer plate and the liquid cultures were incubated in 37° overnight.
