- Ligation mixtures composition:
10 μl digested insert 8 μl digested vector 5 μl Tango buffer(Fermentas) 2 μl ATP 10 mM solution 4 μl MQ water 1 μl ligase T4 (Fermentas)
- Ligation was carried out about 20 hours and subsequently inactivated
