Task 1:

• Prepare the bacterial cultures for isolation of p53 CDS inserted into pSB1A3

Preparation of bacterial cultures:

• Prepare LB medium with kanamycin
• Add 3.5 ml of the medium to the probes
• Add one bacterial colony to each probe
• Breed the bacteria about 7 hours

• Transformation of chemocompetent E. coli strain DH5α

Methods:

• Detailed protocol of transformation is described here. The only modification is usage of total volume of ligation mixture prepared 04.08.09
• petri dish will be held in 37°C for 16 hours
