  • There were no results from last night, since the cells were grown up in the wrong E. Coli.

Clay and Craig

  • There was no growth on the plate from last night's transformations. The wrong strain of E.coli was used. It did not have the death gene resistance.

Tasks

Maria and Afton

  • Last night we had grown up the backbone PSB3K3 in E. Coli Strain NEB10β, however since it contained P1010 we should have used E. Coli strain DB3.1.

Maria and Afton

  • Today we replated cells containing PSB3K3 in E. Coli Strain DB3.1.
  • The controls for the experiment are:
    • E. Coli Cells on LB (+)
    • E. Coli Cells on LB with KAN (-)
    • E. Coli Cells Zapped on LB (+)
